My sister has gone into labor (or at least is dilated enough to merit going to the hospital, I'm a little hazy on strict definitions of when labor starts). Hopefully her daughter will be born today (as opposed to tomorrow...took me 25 hours to be born, so there's family precedent). Updates will be edited in as I get them.

I'm not sure it's a medical definition, but I think it's considered labor once the contractions begin occurring at regular intervals. She could be dilated or effaced close to birth without necessarily being in labor. Her water could even break without her actually being in labor, but if that happens the docs will want to get the baby out soon. More than you wanted to know, I'm sure.
